Recently learning that he has a fatal disease, comic George Simmons (Adam Sandler) spots a struggling performer named Ira (Seth Rogen) and decides to become his mentor. The men become close friends as George teaches Ira the art of stand-up comedy, and Ira helps George define his legacy. When an old flame (Leslie Mann) reappears and the disease goes into remission, George gets a chance to re-evaluate his life and figure out what is truly important to him.

Long Island loser Tommy (Steve Buscemi) is a hopeless alcoholic who loses his job for stealing from his boss, Rob (Anthony LaPaglia), who's now dating his ex, Theresa (Elizabeth Bracco). Tommy copes with his failures by hanging out at local dive Trees Lounge with friends he can barely stand. He takes a gig as the ice cream man, but he's awful with kids. The best thing in his life is a lukewarm romance with cool local girl Debbie (Chloë Sevigny), who, unfortunately, is Theresa's underage niece.

Upper-crust executive Louis Winthorpe III (Dan Aykroyd) and down-and-out hustler Billy Ray Valentine (Eddie Murphy) are the subjects of a bet by successful brokers Mortimer (Don Ameche) and Randolph Duke (Ralph Bellamy). An employee of the Dukes, Winthorpe is framed by the brothers for a crime he didn't commit, with the siblings then installing the street-smart Valentine in his position. When Winthorpe and Valentine uncover the scheme, they set out to turn the tables on the Dukes.

Madison Avenue ad executive Roger (Campbell Scott) attributes his remarkable success with women to his ability to manipulate their emotions from the moment he first meets them, although his confidence is shaken when his boss, Joyce (Isabella Rossellini), unilaterally decides to end their secret workplace affair. When his teenage nephew, Nick (Jesse Eisenberg), drops in for a visit, Roger tries to show him how to pick up women, but soon learns that his approach isn't as foolproof as he thought.

Walter Goodfellow (Rowan Atkinson), vicar of the British village Little Wallop, is busy composing the perfect sermon. Eva has been in charge of her younger sisters ever since their parents died, many years ago. College freshman Whittier (Anne Judson-Yager) and her friend Monica (Faune A. Chambers) make the Cal State cheerleading squad, but can't stand its snobbish leader, Tina (Bree Turner). They resolve to form their own team, pulling together a collection of geeky art students and hippie bleeding hearts whose extracurricular programs were cut for budgetary reasons. Through teamwork, they challenge the official squad's right to represent the school at the upcoming cheerleading championship.
